Hypogonadal Symptoms Are Associated With Different S... [Urology. 2014] - PubMed - NCBI - 0 views

•
This study looked at Total Testosterone and prevalence of low T symptoms. As the authors stated, increased prevalence was found between 320 and 375 ng/dl. Whereas, < 300 ng/dl was consistent with predictable low T symptoms. It would have be more thorough if they had looked at salivary Testosterone levels and bioavailable as well.

Testosterone reduced pain perception in hypogonadism | Endocrine Today - 0 views

•
Study finds that men with opiated induced low T have improvement in pain with Testosterone therapy. Opiates are one of the many causes of low Testosterone (see my book Man Boob Nation). low T has been associated with increase inflammation in men. Thus a normalization of Testosterone would provide anti-inflammatory effects.

Testosterone Substitution Normalizes Elevated Serum Leptin Levels in Hypogonadal Men: The Journal of Clinical Endocrinology & Metabolism: Vol 82, No 8 - 0 views

•
Men with low T and elevated Leptin, have Leptin normalized with Testosterone therapy. In obese men, Leptin is inversely associated with Testosterone. This points to more cause/effect relationship with low T, elevated Leptin, and obesity in men.

Lower sex hormone-binding globulin is more strongly associated with metabolic syndrome than Lower total testosterone in older men: the Health in Men Study - 0 views

•
low SHBG found to be more associated with metabolic syndrome in older men than low Testosterone. The study consisted of 2502 men older than 70. The authors proposed this occurs through the relationship between SHBG and insulin sensitivity. I think that is a little too simplistic.

Inverse relationship between serum levels of interleukin-1beta and testosterone in men with stable coronary artery disease. - ResearchGate - 0 views

•
low Total Testosterone and bioavailable Testosterone is associated with increased IL-1beta and IL-10. This points to low T having a more direct link to increased systemic inflammation in men.

The role of testosterone in the m... [J Steroid Biochem Mol Biol. 2009] - PubMed - NCBI - 0 views

•
Testosterone plays an important role in glucose and lipid metabolism. Thus, low Testosterone, will result in increased disruption in the homeostasis of glucose and lipids resulting in increased weight, insulin resistance, diabetes, metabolic syndrome... low SHBG was also shown to be associated with MetS

Androgen Therapy Improves Insulin Sensitivity and Decreases Leptin Level in Healthy Adult Men With Low Plasma Total Testosterone - 0 views

•
Three month study of men with low T, found that Testosterone therapy improved insulin sensitivity and reduced leptin levels. The group of men here were described as "healthy, non-obese men". Remember, low T is a biomarker of poor health in men, so the authors statement about the study group is inaccurate. This also points to a relatively quick metabolic effect by Testosterone and a direct effect on insulin receptor effect and adipocyte leptin release.

Genetic Determinants of Serum Testosterone Concentrations in Men - 0 views

• mean serum testosterone concentrations were found to be lower in men with GG than in those with TT genotype for rs12150660
• men with the CT genotype for rs6258 had lower serum testosterone concentrations than those with CC genotype.
• The two autosomal SNPs identified by GWAS had a significant influence on the risk of having low serum testosterone (serum testosterone <300 ng/dl) in both the discovery and the replication cohorts with a combined odds ratio (OR) per minor allele of 0.72 (95% CI, 0.65 – 0.79) and 2.7 (95% CI, 2.1 – 3.5) for rs12150660 and rs6258, respectively
• ...3 more annotations...
• The risk of having low serum testosterone concentrations increased by the number of risk alleles with an OR of 1.62 (95% CI, 1.41 – 1.86) for each risk allele (Figure S4). low serum testosterone concentrations were 6.5-times more prevalent in men with ≥3 risk alleles (30.1% prevalence of low serum testosterone) compared to men without any risk allele (4.6% prevalence of low serum testosterone;
• SNP rs5934505 was associated with serum testosterone without SHBG-adjustment (combined p-value of 1.7×10−9) and with free testosterone (combined p-value of 6.7×10−15), but not with SHBG
• The mean serum testosterone and calculated free testosterone but not SHBG concentrations were lower in men with T genotype than in those with C genotype for rs5934505
•
Genetic SNP rs5934505 associated with lower total Testosterone and lower calculated free Testosterone. No effects on SHBG.

Vitamin C increases viral mimicry induced by 5-aza-2′-deoxycytidine | PNAS - 0 views

• Vitamin C alone at concentrations up to 57 μM had little effect on cell growth but was toxic at 228 μM (SI Appendix, Fig. S1B), in line with recent studies of high vitamin C concentrations (125–2,000 μM)
• In our combination approach, vitamin C increased the effects of low doses of 5-aza-CdR, with 57 μM vitamin C almost doubling the growth inhibition
• Using the Chou–Talalay method (28), we found that the two compounds indeed acted synergistically, rather than additively, to inhibit cancer cell growth over the physiological ranges of vitamin C in healthy individuals (26–84 μM)
• ...12 more annotations...
• These results show that targeting the cancer DNA methylome by combining low-dose 5-aza-CdR and vitamin C stimulates the expression of ERVs, the induction of a cell-autonomous immune activation response, and increased apoptosis of cancer cells
• The addition of vitamin C to treatment protocols therefore may be a straightforward way to increase the clinical efficacy of such drugs in MDS and leukemia patients
• Vitamin C deficiency has been seen previously in patients with multiple types of cancer, including hematological malignancies (35⇓–37). We predict that these patients might receive the most benefits from the combination treatment.
• induction of an innate immune response
• We therefore measured plasma concentrations of vitamin C in a small number of patients with miscellaneous hematologic malignancies. Strikingly, 58% of patients with hematological neoplasia who were not taking vitamin C supplements had severe vitamin C deficiency (serum concentration <11.4 μM, at which clinical features of scurvy may be manifested) (34), and 33% had vitamin C levels below the normal range
• it is possible that vitamin C was oxidized to DHA before it was transported into the cells
• Oral administration of vitamin C should be sufficient for the therapeutic strategy, because the concentrations reported in this study would not require i.v. administration.
• Nathan Goodyear

This statement lacks a basic understanding of vitamin C pharmacokinetics.
• Vitamin C is an essential nutrient for humans and has been reported to increase IFN levels in human cells upon virus infection
• daily treatment with vitamin C alone at physiological concentrations enhanced the expression of viral-defense genes relative to untreated cells
• When combined with low-dose 5-aza-CdR, physiological concentrations of vitamin C synergistically inhibited cancer-cell growth and induced apoptosis. Such synergy was associated with increased ERV expression and dsRNA in treated cells. The mechanism of action differs from that of vitamin C at higher doses, which involves its pro-oxidant activity, including GSH inhibition, to generate reactive oxygen species
• This activity has been shown to induce DNA damage and to enhance the sensitivities of myeloid malignancies, multiple myeloma, and cutaneous T-cell lymphoma to arsenic trioxide (41⇓⇓–44). It also can increase chemosensitivity of ovarian cancer cells (27) and selectively kill KRAS or BRAF mutant colorectal cancer cells by inhibiting GAPDH
• reactive oxygen species
•
91% of patients with hematologic malignancies have vitamin C levels that are either low or severly deficient. This study found that vitamin C plus low dose DNA methyltransferase inhibitors have synergistic inhibition of cancer cell proliferation and increased apoptosis. Unfortunately, the authors claimed that oral vitamin C would be sufficient which indicates an incredible lack of understanding of vitamin C pharmacokinetics.
