Progesterone induces the growth and infiltrat... [Biomed Res Int. 2014] - PubMed - NCBI - 0 views

Progesterone shown to stimulate growth and migration of astrocytes in the brain.  The has both implications in cancer, such as in astrocytomas, but also in TBI.  The study also found that the Progesterone antagonist RU486 inhibited this effect.  A more thorough evaluation would have included the pregnane metabolites.

Plasma Membrane Receptors for the Cancer-Regulating Progesterone Metabolites, 5α-Pregnane-3,20-dione and 3α-Hydroxy-4-pregnen-20-one in MCF-7 Breast Cancer Cells - 0 views

  •
The receptor for the progesterone 5alpha pregnane, 5alpha-P, is unregulated in breast cancer cell line MCF-7.  Estradiol increases the receptor expression.  Not only is there an enzymatic effect in these progesterone pathways, but the receptor is equally unregulated.

Membrane 5α-pregnane-3,20-dione (5αP) receptors in MCF-7 and MCF-10A breast cancer cells are up-regulated by estradiol and 5αP and down-regulated by the progesterone metabolites, 3α-dihydroprogesterone and 20α-dihydroprogesterone, with associated changes - 0 views

Very interesting study.  In ER/PR + breast cancer cells, Estradiol and 5alpha-dihydroprogestorone increase the membrane expression of 5alpha-dihydroprogesterone receptors.  This has a pro-growth, pro-metastatic potential.  In contrast, the metabolites 3alpha-dihdroprogesterone and 20alpha-HP inhibit the same membrane receptor expression thus giving an antigrowth, anti-metastatic potential.

knowledge of progesterone and its metabolites are important in the use of hormones.  As the authors concluded here: "natural steroids should not be disparaged...an update on endocrinological knowledge and experience is rather mandatory for gynecologists".  There in lies the problem.  The IOM has shown that the average physician practices at a level of 17 years behind the current scientific knowledge and the environment created by medical governing bodies discourages questions, so as to protect the system.

Progesterone-induced stimulation of mammary tumorigenesis is due to the progesterone metabolite, 5α-dihydroprogesterone (5αP) and can be suppressed by the 5α-reductase inhibitor, finasteride - ScienceDirect - 0 views

Membrane 5alpha-pregnane-3,20-dio... [J Steroid Biochem Mol Biol. 2005] - PubMed - NCBI - 0 views

Progesterone metabolites are present on the membrane of ER/PR + as well as ER/PR - breast cancer cell lines.  The balance of 5 alpha pregnane:3 alpha pregnane regulates tumor genesis.  Estradiol increased 5 alpha pregnane receptors as well as 5 alpha pregnane unregulated itself via autocrine activity.  The 3 alpha pregnenes, 3 alpha hydroxyprogesterone and 20 alpha-dihydroprogesterone, decrease the 5 alpha pregnane receptors via paracrine activity.

The progesterone metabolite allopregnanolone is shown to play a significant role in the PMDD symptoms.  The mechanism is via a paradoxical GABA-A effect.

Activity and expression of progesterone metabolizing 5α-reductase, 20α-hydroxysteroid oxidoreductase and 3α(β)-hydroxysteroid oxidoreductases in tumorigenic (MCF7, MDA-MB-231, T-47D) and nontumorigenic (MCF10A) human breast cancer cells - ResearchGate - 0 views

Intra mammary progesterone metabolism and its associated metabolites found to be associated with tumorigenic activity in cell lines.  This was independent from ER PR status.  5 alpha-pregnanes were found to be pro-tumor and 4-pregnanes were anti-tumor.

Might the enzymes be more important than the hormones themselves?  The number is important i.e. progesterone, but how the number came to be is more important as it points to the underlying etiology.  Here, the balance of enzymes are found to direct cell line tumor potential.  The enzyme 5alpha-reductase increased production of the 5alpha-pregnanes which increased tumor potential.  In contrast, the enzymes 20alpha-hydrosteroid oxidoreductase and 3alpha-hydroxysteroid oxidoreductase increase production of the 4alpha-pregnanes, which decreased tumor potential.

Breast cell culture study finds increase 5alpha-reductase 1/2 activity and decreased 3alpha HSO.  This fits previous previous studies that show that increased 5alpha pregnanes is associated with increased proliferation and increased detachment; in contrast to decreased 4-pregnene metabolites.

20alpha dihydroprogesterone, a 4 pregnene, has been shown to inhibit breast cancer cell proliferation, decrease mets through increased cell adhesion, and increased apoptosis.  Here, it has been shown to inhibit aromatase activity.  No longer are hormone metabolites to be viewed as waste.  They are biological as active if not more so in many cases.

Just the abstract available here.  Article states the complexity of hormones found in hormone metabolism.  I love the authors quote, "As steroid hormones are known to be converted to many other steroids occupying different receptors and thereby exerting various different effects..."  Doesn't fit into the "know your numbers" marketing deception does it?
