Overexpression of SIRT1 Protects Pancreatic β-Cells Against Cytokine Toxicity... - 0 views

  • SIRT1, a class III histone/protein deacetylase, is known to interfere with the nuclear factor-κB (NF-κB) signaling pathway and thereby has an anti-inflammatory function
  • central role of NF-κB in cytokine-mediated pancreatic β-cell damage
  • SIRT1 as a possible target to attenuate cytokine-induced β-cell damage.
  •
SIRT1 as possible target to reduce pancreatic inflammation and thus prevent/reduce diabetes;  Oh by the way Resveratrol acts at SIRT1

Neuroprotective Sirtuin ratio reversed by ApoE4 - 0 views

  •
ApoE4 allele is known to increase Alzheimer's disease by 10 fold.  One mechanism is through a lowering of SirT1 and an increase in the SirT1:SirT2 ratio.  This SirT1 is the target of resveratrol in red wine.

SIRT1 Mediates Central Circadian Control in the SCN by a Mechanism that Decays with Aging - 0 views

  •
SIRT1, known to reduce inflammatory signaling, found to help maintain circadian rhythm.  Loss of the circadian rhythm is know to contribute to chronic disease.  Likewise, SIRT1 has been shown to reduce inflammation, which will reduce disease.  Is this their link?

Evidence for a Common Mechanism of SIRT1 Regulation by Allosteric Activators - 0 views

  •
science validates the health benefits of resveratrol.  The benefit is through SIRT1 activation.

Mechanism of Human SIRT1 Activation by Resveratrol - 0 views

  • The NAD+-dependent protein deacetylase family, Sir2 (or sirtuins), is important for many cellular processes including gene silencing, regulation of p53, fatty acid metabolism, cell cycle regulation, and life span extension
  • resveratrol was shown to increase life span in three model organisms through a Sir2-dependent pathway.
  •
Resveratrol potentially increases life span through SIRT1 activation

Nitic Oxide (NO) via inflammation deactivates SIRT1 is a common finding in inflammatory diseases of aging, proposes authors of this paper.

Declining NAD+ Induces a Pseudohypoxic State Disrupting Nuclear-Mitochondrial Communica... - 0 views

  •
NAD found to slow mitochondrial decline associated with aging.  SIRT1 is involved; interesting of note is the inverse association with HIF-1alpha

The NAD+ precursor nicotinamide riboside enhances oxidative metabolism and protects aga... - 0 views

  •
nicotinamide riboside shown to offset the negative metabolic effects of American diet induced obesity.  NAD also increased SIRT1 and SIRT3 activity

Modulation of NF-[kappa]B-dependent transcription and cell survival by the SIRT1 deacet... - 0 views

  • Treatment of cells with resveratrol, a small-molecule agonist of Sirtuin activity, potentiates chromatin-associated SIRT1 protein on the cIAP-2 promoter region, an effect that correlates with a loss of NF-B-regulated gene expression
  •
resveratrol decreases NF-kappa Beta expression and thus reduces inflammation

THE NEUROPROTECTIVE PROPERTIES OF CALORIE RESTRICTION, THE KETOGENIC DIET, AND KETONE B... - 0 views

  •
Fantastic read on the biochemical benefits of calorie restriction and a ketogenic diet.

Is Timing Everything? New Insights into Why the Effect of Estrogen Therapy on Memory Mi... - 0 views

  • Women who have an oophorectomy before the normal age at menopause show an increased risk for cognitive impairment or dementia later in life unless they are treated with estrogen until the normal age at menopause
  • SIRT1 has been implicated in the disruption of mitochondrial bioenergenetics in Alzheimer's disease and mild cognitive impairment
  • the increase in dementia observed with CEE/MPA rather than CEE alone suggests potential deleterious effects of MPA on brain function in older women
  • ...1 more annotation...
  • SIRT 1 as a potential mediator of the impact of E2
  •
Early estrogen therapy in perimenopause and early menopause, with Estradiol, provides more health benefits than later therapy.  This article looked at Estrogen's effects on a woman's brain.  This likely has its origins in the change in estrogen receptors. The signal is not changing, but the reception of that signal is.  How else can one explain a different response to the same hormone dosage?
