In this episode of Think Like A Scientist, we meet conservation photographer Krista Schlyer, who has spent the last seven years documenting the environmental effects of the U.S./Mexico border wall, and biologist Jon Beckmann, who studies how man-made barriers influence the movement of wildlife. Schlyer and Beckmann have seen damaging impacts of the border wall firsthand, but they remain optimistic. Humans probably won't stop constructing walls and fences any time soon, but planning our boundaries with wildlife in mind can help prevent these structures from causing environmental harm.

    Women who are overweight while pregnant are more likely to have babies who are biologically older than those born to women of a healthy weight. This could put the babies at a higher risk of developing chronic diseases later in life, and may reduce their life expectancy. Our biological age is linked to the length of our telomeres - bits of DNA that cap the ends of our chromosomes. Our telomeres shrink every time our cells divide, and continue to shorten throughout life. "Short telomeres have been associated with cardiovascular disease, type 2 diabetes and atherosclerosis," says Tim Nawrot at Hasselt University in Belgium.

    This interrupted case study examines molecular genetic evidence reported in scientific literature to determine the fate of Louis-Charles, son of Louis XVI and Marie-Antoinette of France. Controversy and rumors surrounding the death of Louis-Charles suggested that either he died as a young boy while being held in captivity by the French revolutionaries or he escaped and was replaced by a substitute who died in his place. One individual claiming to be Louis-Charles was Karl Naundorff. Students begin the case by preparing pedigrees for the descendants of Maria Theresa and Francis I, the Holy Roman Emperor, parents of Marie-Antoinette. The pedigrees can be used to introduce the concepts of alleles identical-by-descent and cytoplasmic inheritance patterns. Students then compare mitochondrial DNA sequences and XY chromosome sequences from hair, bone, heart, and blood samples taken from descendants of Marie Theresa, Karl Naundorff and the heart of the boy who died in captivity to determine if the latter was truly Louis-Charles. An optional PowerPoint presentation with clicker questions is available to help guide the classroom activities.

    In a retrospective cohort study investigating the role of epigenetic control in the onset of allergic diseases, it was shown that mothers who suffered from allergic symptoms very early in pregnancy were more likely to have offspring with allergies. This was compared to mothers who did not suffer from allergies in early pregnancy, taking into account family history of allergies and total allergic response during pregnancy (10). These results indicate that gene-environment interactions during embryonic development may modify the epigenetic code in ways that may potentially lead to allergic disease.

    "This modular case study tells the story of Dan and Annie, a married couple of Acadian ancestry who have a genetic form of deafblindness called Usher syndrome. They live in Southwest Louisiana, home of the largest population of DeafBlind citizens in the United States. Acadian Usher syndrome is caused by an allele of the USH1C gene that came to Louisiana with the first Acadian settlers from Canada who founded today's Cajun population. This allele's single nucleotide substitution creates an erroneous splice site that produces a defective cytoskeletal protein (harmonin) of the cochlear and vestibular hair cells and retinal photoreceptors. This splice site is the target of a promising gene therapy. The case study applies and connects Mendelian inheritance, chromosomes, cell division, vision and hearing, DNA sequences, gene expression, gene therapy and population genetics to a specific gene and its movement through generations of Dan and Annie's families.  After the introduction, each of the remaining sections can be used independently either for in-class team activities or out-of-class extensions or assignments over an entire year of introductory undergraduate biology. "

    This case study describes a visit by "Naomi" to her physician, who upon seeing Naomi's bloodwork decides to prescribe her a cholesterol-lowering agent, a statin. The case discusses (1) the prevalence of non-communicable diseases such as atherosclerosis, which can lead to strokes and heart disease; (2) circulating lipoprotein particles including LDL and HDL, differentiating between which is a risk factor for heart disease and which is protective;  (3) the pathway for cholesterol synthesis; (4) the evidence demonstrating that statins inhibit the enzyme HMG-CoA reductase, which is the committed step for cholesterol synthesis; and (5) the kinetic mechanism by which statins inhibit the enzyme. Students examine and interpret data from radioactive tracer and enzyme kinetics studies. The case ends with another visit by Naomi to her physician, who reexamines her bloodwork after she has been taking a statin for six months. Students are asked to determine whether the drug was effective for Naomi and, if not, what changes could be made to her regimen. The case was originally written for use in a survey of biochemistry course for upper level undergraduates.

    This case study was written for an introductory course for biology majors who are first learning about embryonic development. The case is composed of several parts and involves a storyline about a team of researchers who find frogs and eggs in bamboo plants during a field study. Students consider what these observations mean, learn basics about the stages of animal embryonic development, and make connections to phylogeny and natural selection. Students then apply their understanding of animal embryonic stage development to the chemical atrazine in the environment by examining data from several experiments. As a concluding activity students write a letter to an agency or newspaper of their choosing stating their opinion surrounding the use of atrazine in the environment. The case proceeds in a progressive disclosure format and involves a combination of class discussion, small group work, and homework. Because the case focuses on very basic animal embryonic development, it would also be a great start to a developmental biology course or an embryology course.

    That's why a paper published Thursday in the journal Genetics in Medicine is so remarkable. The paper identifies NGLY1 deficiency as an inherited genetic disorder, caused by mutations in the NGLY1 gene. The researchers have confirmed eight patients with these mutations who share several symptoms, including developmental delays, abnormal tear production and liver disease.
