Create a translational science network devoted exclusively to immunotherapy approaches to treat and prevent adult cancers.  The purpose of this FOA is to develop Immuno-engineering to Improve immunotherapy (i3) Centers, as new components of the Immuno-Oncology Translation Network (IOTN). The i3 Centers will be comprised of multi-disciplinary teams focused on developing and employing engineered immunotherapy approaches to design more durable, widely accessible, and less toxic immunoprevention and immunotherapy strategies. Advancing Cancer Immunotherapy by Mitigating Immune-related Adverse Events (irAEs) (U01 Clinical Trial Not Allowed) - 0 views

    This FOA will invite U01 applications in the specific area of cancer immunotherapy-related adverse events. Investigators are encouraged to submit applications that focus on mechanisms of immune reactivity/tolerance and/or autoimmune pathways that could be applied to improving immunotherapeutic approaches while simultaneously eliminating or reducing the severity of inflammatory or autoimmune responses. A related area of interest is studies designed to enhance the target specificity of immunotherapeutic reagents, to reduce or prevent irAEs. An additional synergistic focus of interest is the identification of predictive biomarkers of cancer patients at risk for developing irAEs. Understanding risk factors for developing an irAE would better inform patient stratification at the start of therapy and influence clinical monitoring. Achieving the goals of the RFA should establish a deeper understanding of the origins and activation pathways leading to inflammatory or autoimmune adverse events that currently limit the use of various immunotherapy regimens in patients.

    Significant advances in the understanding of the pathophysiology and molecular biology of multiple myeloma have identified numerous molecular targets for therapeutic intervention. Immunotherapeutic approaches (e.g., Daratumumab and Elotuzumab) have established themselves in the armamentarium for multiple myeloma, and ongoing adoptive cell therapy trials provide encouraging signs of clinical activity. As immune-based therapeutic agents continue to advance their way through the clinic, there is a critical need for increased understanding of the role that the immune system plays in myeloma disease biology, disease progression, and therapeutic response and resistance. The knowledge developed through this RFP will be critical to fully developing precision immunotherapy for the treatment of myeloma.

    The purpose of this Funding Opportunity Announcement (FOA) is to accelerate the adoption and validation of molecular/cellular/imaging markers and assays for cancer detection, diagnosis, prognosis, monitoring, and prediction of response or resistance to treatment, as well as markers for cancer prevention and control.     To that end, the two organizations have issued a Request for Applications for projects that test feasibility and generate preliminary data. MRA and ACS have each committed $1 million for the awards. The combined $2 million will be used to fund Multidisciplinary Team Awards and Pilot Awards. 1) Multidisciplinary Team Awards: At least one grant of up to $1 million will be awarded in this category are designed to foster interdisciplinary collaborative research to promote transformational advances with the potential for rapid clinical benefit. Letters of Intent must be received no later than August. Upon review, selected applicants will be invited to submit full applications by October 31, 2017. 2) Pilot Awards: Up to five grants of up to $200,000 each will be awarded over two years to help seed the ground in this relatively young area of checkpoint irAE research. Applications must be received no later than October 16, 2017.
