    Because the gene-enzyme system forms a closed loop, it presents us with a classic chicken or egg conundrum: Which came first, genes or the protein enzymes they code for? While the details are still not fully worked out, discoveries over the past few decades have lead researchers to a surprising possible solution: What really came first? Genes that act as enzymes! The RNA World Hypothesis is the idea that before living cells, the genetic code, and the gene/protein cycle ever existed, chains of a chemical called RNA were forming naturally. Once formed, some of these chains were able to function as enzymes, and were even able to evolve by making copies of themselves with slight, accidental modifications.

    This clicker case was designed to teach students about basic enzyme structure, mechanisms of enzyme inhibition, and mechanisms of drug resistance. The story follows Oliver Casey, a patient afflicted with Chronic Myelogenous Leukemia (CML). CML is caused by a chromosomal mutation that affects the tyrosine kinase ABL, an enzyme important in regulating cell growth and proliferation. The chromosomal mutation gives rise to the BCR-ABL fusion gene that produces a constitutively active ABL kinase, which causes the leukemia. In May 2001, the Food and Drug Administration approved the use of a rationally designed tyrosine kinase inhibitor, imatinib (Gleevec®), for the treatment of CML. During that same month, Gleevec made the cover of TIME magazine, described as "new ammunition in the war on cancer." The case is structured for a flipped classroom environment in which students view preparatory videos (including one by the author) on their own before beginning the case. Written for a first-year introductory biology course, the case could also be adapted for AP/Honors high school biology or a cancer biology course.

    This case study describes a visit by "Naomi" to her physician, who upon seeing Naomi's bloodwork decides to prescribe her a cholesterol-lowering agent, a statin. The case discusses (1) the prevalence of non-communicable diseases such as atherosclerosis, which can lead to strokes and heart disease; (2) circulating lipoprotein particles including LDL and HDL, differentiating between which is a risk factor for heart disease and which is protective;  (3) the pathway for cholesterol synthesis; (4) the evidence demonstrating that statins inhibit the enzyme HMG-CoA reductase, which is the committed step for cholesterol synthesis; and (5) the kinetic mechanism by which statins inhibit the enzyme. Students examine and interpret data from radioactive tracer and enzyme kinetics studies. The case ends with another visit by Naomi to her physician, who reexamines her bloodwork after she has been taking a statin for six months. Students are asked to determine whether the drug was effective for Naomi and, if not, what changes could be made to her regimen. The case was originally written for use in a survey of biochemistry course for upper level undergraduates.

This gene codes for a protein that simply places a "routing tag" on certain enzymes that shred cellular trash. The tag ensures that the shredding enzymes end up in chambers called lysosomes, basically the cell's garbage disposal. It's a mundane cellular activity, yet mutations in the same process in humans have also been linked to stuttering-a bizarrely specific condition for such a general gene.     In the study, published Monday (May 1) in Nature Biotechnology,  Luo and colleagues set their sights on two fusions genes they had previously found to be associated with prostate cancer and various forms of rapid and invasive cancer, including liver tumors. Using a modified CRISPR-Cas9 tool that creates a single- rather than double-stranded break in DNA, they targeted the chromosomal breakpoints that form these fusion genes and replaced fusion DNA with a gene encoding the enzyme HSV1-tk. This enzyme effectively kills tumor cells by converting the drug ganciclovir into its active form, which then blocks DNA synthesis and leads to cell death. (Ganciclovir is used to treat cytomegalovirus in humans.)
