Single Cell Opioid Responses in the Context of HIV (SCORCH) Program: Data Coordination, Analysis, and Scientific Outreach (UM1 Clinical Trial Not Allowed) - 0 views

    Along with RFA-DA-19-037, which will support the generation of single cell datasets for one or more brain regions relevant to opioid use disorder and persistent HIV infection, a data coordination/analysis/outreach center (RFA-DA-19-038) will be established to make NIDA-funded single cell data and other molecular HIV/SUD data FAIR (Findable, Accessible, Interoperable, and Reusable). Harmonized single cell HIV/SUD data sets will enable immediate data mining by the scientific community for HIV and/or SUD biomarkers and potential pathways for therapeutic intervention. It will also enable future mining of these data sets as new and improved data science and information technology approaches are developed, maximizing NIDAs original investment in the data generating activities.

Rare Disease Cohorts in Heart, Lung, Blood and Sleep Disorders (UG3/UH3 Clinical Trial Not Allowed) - 0 views

    The purpose of this FOA is to fund research centers that will establish longitudinal cohorts in rare HLBS diseases to investigate unaddressed research questions using epidemiologic study designs and methods that are appropriate for conditions affecting fewer than 200,000 persons in the US. These observational cohort studies should be designed to provide an evidence base for future interventional studies, including clinical trials; for developing better diagnostics than those that are currently available; for answering early translational questions; or for broader implementation of guidelines for managing these diseases. This program will provide opportunities to advance rare disease research using genetics and deep phenotyping to characterize the disease and to identify disease sub-types; to use data science methods that integrate clinical and patient-reported outcomes (PROs) with laboratory, imaging, environmental and -omics data to understand the natural history of disease; to generate data that differentiate patients with the same morphological phenotype but different genetic mutations and severity of outcomes; to elucidate genotype-phenotype interactions and multisystem phenotyping to develop reliable and valid predictive tools to determine who will respond to which treatments and when to intervene; and to encourage innovative methods such as telemedicine to include participants with rare diseases located in remote locations.

Qualitative research is a form of social inquiry that seeks to understand the human experience by exploring how people interpret and make sense of their experiences and the world in which they live.     Supported by the NIH Common Fund (Common Fund) Science of Behavior Change (SOBC) Program, this Funding Opportunity Announcement (FOA) solicits competitive revision (formerly known as a competitive supplement) applications to NIH-supported clinical trials awarded as research project R34 grants. The goal of the SOBC Program is to advance a mechanisms-focused, experimental medicine approach to behavior change research. Funded projects in the SOBC Research Network (https://commonfund.nih.gov/behaviorchange/fundedresearch) have developed experimental manipulations, assays, and/or measures (hereafter referred to as assays for brevity) to support an experimental medicine approach to behavior change research. The SOBC Measures Repository is accessible from the SOBC Research Network Open Science Framework (OSF) page at https://osf.io/zp7b4. The goal of this FOA is to accelerate the adaptation, validation, and translation of SOBC Research Network assays for use in ongoing clinical trials. This FOA calls for the integration of SOBC Research Network assays into active NIH-supported clinical trials of drugs, devices, procedures, or behavior modifications. As such, the active NIH-supported clinical trial used to respond to this FOA does not have to be a behavior change trial or identify behavior change as a primary outcome. The integration of SOBC Research Network assays into ongoing clinical trials will accelerate the development of interventions and experimental manipulations that have been shown to engage specific mechanisms of behavior change and the development of assays that verify engagement of those behavior change targets.

    This FOA invites applications that propose the comprehensive functional validation of newly identified therapeutic target candidates for Alzheimer's Disease-Related Dementias (ADRDs). This FOA seeks to promote critical target validation approaches to help de-risk subsequent translational research and accelerate the advancement of novel therapies for ADRD. Target(s) or molecular pathway(s) to be considered for validation must have been already identified using tissue expression or genetic data generated in human samples. In its initial phase, this FOA provided support for up to two years (UG3 stage) for the development of customized technologies, models, and protocols to modulate the expression or activity of target candidate(s) in cells or tissues and monitor their functional biological consequences in in vitro or in vivo disease models. Upon demonstration of technical feasibilities, a second phase (UH3 stage) will carefully and reproducibly measure and cross-validate the impact of the target modulation in different modalities across collaborating laboratories using the NIH rigor and reproducibility guidelines. Applicants responding to this FOA must address objectives for both the UG3 and UH3 phases and are expected to have a substantial collaborative effort between independents laboratories. This FOA is not specific for any one or group within the ADRD spectrum of disorders. Disorders covered in these applications are frontotemporal degeneration (FTD), Lewy body dementias (LBD) (including dementia with Lewy bodies (DLB)), Parkinson disease dementia (PDD), vascular contributions to cognitive impairment and dementia (VCID), mixed dementias including the associated diagnostic challenges of multiples etiology dementias (MED).

RFA-NS-19-026: Clinical and Biological Measures of TBI-related dementia including Chronic Traumatic Encephalopathy (CTE) (R01 Clinical Trial Not Allowed) - 0 views

    This FOA invites investigation of biological and clinical measures of TBI-related progressive neurodegeneration and neurocognitive decline associated with increased risk for dementia and /or traumatic encephalopathy syndrome (TES) (clinicopathologic diagnostic counterpart to the neuropathological diagnosis of Chronic Traumatic Encephalopathy (CTE)). Investigations should be based on existing, well-characterized populations of patients with a history of TBI that are enriched for increased risk of cognitive impairment or dementia and can continue to be followed longitudinally; additional subjects may be recruited as appropriate. The overall goal is to advance knowledge of the underlying pathophysiology and clinical characterization of the chronic effects of TBI that distinguish static-chronic TBI cognitive impairment from those that lead to progressive neurodegeneration associated with TES and dementia. A critical feature of this FOA includes the broad sharing of clinical, neuroimaging, physiological, and biospecimen data to further advance research in this area

Towards Implementing Novel Training Methods to Enhance Cognition in Aging (U01 Clinic - 0 views

    This RFA invites applications for planning awards to develop and finalize protocols for well-powered cognitive training intervention trials to remediate or prevent age-related cognitive decline as well as possibly prevent or delay the onset of mild cognitive impairment and dementia. Planning activities may include the collection of pilot data and the refinement of cognitive training protocols consistent with Stage I of the NIH Stage Model. Trial designs must justify the means used to assess cognition and to explore the underlying mechanisms of change. Such methods as structural and functional neuroimaging with biomarkers justified by an underlying model of change, CSF fluids, and blood biomarkers are appropriate candidate tools.
