    Despite established associations between white matter lesions and cognitive impairment including dementia, little prospective information is available to define the characteristics of white matter lesions and associated comorbid clinical factors that cause further cognitive impairment, ultimately resulting in dementia. This initiative will support one large prospective clinical research study in the U.S. of patients engaged with the health care system because of incidental white matter lesions found on neuroimaging, who present with cognitive complaints, and who thus are at risk for cognitive decline. The goal will be to determine the volume and anatomical features that are both necessary and sufficient to cause cognitive impairment or dementia.

    This initiative will provide two years of support for planning activities necessary for initiating a Phase III clinical trial designed to validate VCID biomarkers and/or treat patients with VCID. The full spectrum of VCID is in scope, and the one or more VCID disorders(s) to be targeted in the trial must be clearly specified in the application, for example (but not limited to): vascular insults including clinical stroke, silent infarcts and microinfarcts, leukoaraiosis, cerebral amyloid angiopathy (CAA), transient ischemic attack (TIA), micro-bleeds, CADASIL, and vascular contributions to Alzheimers dementia.     The goal of this FOA is to support basic and clinical research on the influence of alcohol on susceptibility and progression of Alzheimer's disease and its related dementias. Recent longitudinal studies have provided strong evidence that alcohol use disorder is associated with the high risk of all types of dementias, and frequent heavy drinking increases risk of both Alzheimer's disease and vascular dementia. Even moderate alcohol consumption may be a risk factor for adverse brain outcomes and cognitive decline. Although these studies link heavy and frequent alcohol drinking to dementias in aging populations, mechanisms contributing to this relationship are not well understood. With this FOA, we solicit research projects that combine diverse expertise and use innovative approaches to investigate mechanisms by which alcohol affects brain aging processes to produce dementias and influences development of Alzheimer's disease. This FOA strongly encourages collaborations between alcohol researchers and experts in Alzheimer's disease and its related dementia research

    CZI seeks applications aimed toward identifying unifying principles that underlie tissue homeostasis and inflammation at the single cell level. Our goal is to stimulate collaborations across disciplines that will help define a new field. Successful applications will bring together researchers in different experimental, computational, or medical domains. They will address local cell properties and interactions in inflamed tissues and compare them to the properties and interactions of similar cells in healthy tissues. They will increase our understanding of the cell types that mediate inflammation, and their interactions in space and time. The two-year pilot grant period is intended to develop proof-of-concept for the experimental team and the approach, setting up future programs for detailed mechanistic investigations.
