As exogenous GABA cannot penetrate the blood-brain barrier, it is synthesized in the GABAergic neurons in the CNS, converted from glutamate, the principal excitatory neurotransmitter, using the enzyme glutamic acid decarboxylase (GAD) and pyridoxal phosphate as a cofactor.
