WHAT IS PROTEINASE K MADE OF? Proteinase K is known enzyme belonging to the class of serine proteases that cleave adjacent to the carboxylic group peptide bonds of aromatic and aliphatic amino acids. It's also exhibited a very broad cleavage specificity. The enzyme was discovered in extracts of the fungus Engyodontium album. HOW DOES PROTEINASE K WORK? In molecular biology, main application of proteinase K is to digest proteins during nucleic acids purification in order to remove protein contaminants: - Proteinase K quickly inactivates nucleases (enzymes that digest nucleic acids) that degrade DNA or RNA molecules during the cleansing. - Proteinase K is activated by calcium. It does not affect the activity of the proteinase, but protects again autolysis, improves thermal stability, and regulates substrate binding capacity. To increase the stability of Proteinase K, thereby enhancing its action, Urea as well as SDS (sodium dodecyl sulfate) or elevated temperature at 37-60C is commonly used. Inactivation of Proteinase K is occurring at temperature above 65C. WHAT IS THE PURPOSE OF PROTEINASES? The General Pharmaceutical Council (GPhC) has published a new strategy pledging to deliver equality, improve diversity and foster inclusion in all aspects of its work. The strategy was updated after a public consultation held between April and July. It's key themes are: To make regulatory decisions that are demonstrably fair, lawful, and free from discrimination and bias. To use our standards to proactively help tackle discrimination and to make sure everyone can access person-centred care, fostering equality of health outcomes.To lead by example and demonstrate best practice within our organisation, holding ourselves to the same high standards we expect of others. GPhC chair Nigel Clarke said having a strategy is vital as the pandemic has exposed "the scope and scale of inequities in society, in healthcare outcomes and in pharmacy." "Inequality and exclusion are bad for people's health. Therefore, this strategy is fundamental to our core purpose as a regulator and our vision for safe and effective pharmacy care at the heart of healthier communities."

The Association of Independent Multiple Pharmacies (AIMp) has called upon all pharmacy organisations to unite and collectively represent the sector. While underlining the need for the sector to adopt a straightforward manifesto that can be presented to the public, politicians, and decision-makers, the association also advocated for a revitalisation of the 'Save Our Pharmacies' campaign, injecting it with renewed energy and a stronger sense of purpose. AIMp pointed out that a lack of cohesion is one of the factors contributing to the sector's struggle in gaining attention. "The current trajectory of community pharmacy is unsustainable," said Dr. Leyla Hannbeck, Chief Executive of AIMp. "To tackle the significant challenges ahead, we must work together cohesively. The most effective approach is to actively engage with Parliament, leveraging politicians from across the political spectrum to advocate for our cause." Meanwhile, the association urged various pharmacy bodies to unite in support of the Pharmacy All Party Parliamentary Group (APPG) to enhance its effectiveness and broaden its influence, rather than working independently.

The General Pharmaceutical Council (GPhC) has urged health and care professionals to meet relevant regulatory standards amidst the shortage of GLP-1 receptor agonists (GLP-1 RAs). The council said: "We are concerned to hear that people with Type 2 diabetes are experiencing problems accessing GLP-1 receptor agonists (GLP-1 RAs). We know that these are being widely prescribed for weight loss purposes and in some cases prescribed off-label." "We recognise the adverse impact that shortages and supply chain issues can have on patients, the public and wider health and care teams. The Council understands that health and care professionals are using their professional judgement and making decisions in challenging situations, balancing a range of factors such as individual patient needs, wider public health and pressures and limitations on available resources such as medicines shortages or other supply chain issues.

The Royal Pharmaceutical Society (RPS) has published a consultation to review the 'Professional standards for the reporting, learning, sharing, taking action and review of incidents error reporting' in collaboration with Association of Pharmacy Technicians UK and Pharmacy Forum of Northern Ireland. The consultation is open from 21 April 2023 until 2 June 2023 and are due to be published in early 2024. All three professional leadership bodies are calling for contributions from anyone using the standards to ensure they are clear, current, relevant and fit for purpose. "The review affects all pharmacists, pharmacy technicians, trainees and members of the pharmacy team working in all sectors across the UK," RPS said. "The standards may also be of interest to the public, people who use pharmacy and healthcare services, healthcare and professionals working with pharmacy teams.

The latest inspection report of the General Pharmaceutical Council (GPhC) revealed that nine pharmacies did not meet their standards out of 52 pharmacies that were inspected between January 2023 and May 2023. Pharmacies are inspected on five principles - Governance, Staff, Premises, Services including medicines management and Equipment and facilities. Out of nine, eight pharmacies did not meet GPhC standard of governance which is defined to safeguard the health, safety and wellbeing of patients and the public. Six pharmacies did not meet the GPhC standard of principle four which defines the way in which pharmacy services, including the management of medicines and medical devices, are delivered safeguards the health, safety and wellbeing of patients and the public. The purpose of these standards is to create and maintain the right environment, both organisational and physical, for the safe and effective practice of pharmacy. The standards apply to all pharmacies registered with GPhC.

A winter NHS crisis is inevitable unless the government acts now to reverse the worrying decline in community pharmacies. Years of government underfunding could see 3,000 pharmacies in England - around a third of the network - having no option but to shut their doors to patients in the next few years. That figure is based on independent assessments from Ernst & Young and UCL/LSE healthcare professors: it is not scaremongering - it is the reality the country faces. Fifty per cent of pharmacies are already in financial distress because government funding has been falling in real terms since 2019 and that figure is predicted to rise to 75 per cent within the next two years. The government needs to act now and invest in pharmacy or sleepwalk into a healthcare disaster as we have seen with access to dentistry care. Prescription volumes have risen consistently year-on-year by roughly 2 per cent which means fewer pharmacies doing more work and under greater pressure than a decade ago. Ten years ago around 11,200 pharmacies in England were dispensing roughly 79,000 prescriptions; nowadays around 11,500 are dispensing roughly 89,000 prescriptions. The secretary of state recently asked pharmacy to do more to avoid a winter NHS crisis and at the same time said there will be no new money to pay for those additional services. This at a time when the network is in decline with random unplanned pharmacy closures - 640 closures since 2016 - and pharmacy staff face huge workload pressures as prescription demand is increasing year-on-year. The government's approach to pharmacy literally does not add up: the pharmacy contract is not fit-for-purpose now let alone dealing with a NHS winter crisis.

The Royal Pharmaceutical Society (RPS) and the charity Marie Curie have developed a consultation on 'professional standards for palliative and end of life care for community pharmacy'. The consultation will be open for four weeks for community pharmacists, organisations with a role in community pharmacy or palliative/end of life care, or patients or carers with first-hand experience of palliative and end of life care to share their views until 1st December 2022. Through this consultation, Society aims to that ensure the standards are clear, relevant, current, and fit for purpose from the perspective of the community pharmacy team, the wider health team and patients/carers. "The final standards will be published in 2023 and will help community pharmacies across the UK to self-assess and continuously improve their palliative, end of life and bereavement care," said RPS.
